Abstract

|
To evaluate the expression level of extracellular superoxide dismutase (EC-SOD) in various tissues, we purified EC-SOD from skin, lung, brain, kidney, liver and spleen of 4 and 8 weeks old FVB/NJ mice and measured its activity. EC-SOD activity in the skin was 10- to 19-fold higher than other tissues. This suggests that EC-SOD may be expressed massively in the skin and this enzyme may play an important role in the skin. Expressions of EC-SOD gene in vectors-transfected Pam212 cells were over two times higher than in control in vitro analysis. And EC-SOD activities in the cultured media of vectors-transfected cells were 5- to 6-fold higher than that of control. These results suggest that the vectors may not only induce keratinocyte-specific expression of EC-SOD but also express successfully functional EC-SOD.
